- add the sample and primers to the Isothermal Master Mix
- amplify at a constant temperature of 65 °C for isothermal conditions (even a heat block and/or waterbath can be used for this! No need to buy an expensive thermal cycler)
- Fluorescence detection can be done on either gel electrophoresis, on our Genie II or Genie III LAMP instruments in real time, or visually with the naked eye thanks to colour change by the intercalating dye included in the Isothermal Master Mix, which also allows for a complete, closed-tube system
The Genie II LAMP system (pictured above) is a sophisticated, portable instrument which can perform isothermal amplification of DNA and RNA in minutes, and display fluorescent results in real time on a 7" LCD touchscreen. The system uses an anneal curve to confirm molecular results. Genie II comes fitted with a rechargeable Li-Po battery which allows for operation during power outages or out in the field.
We also have a younger sister to the Genie II, that would be the Genie III Point-of-Application LAMP portable system (pictured below) which can be used out in the field to get quick isothermal molecular results. It's weather-proof, has a touchscreen, rechargeable battery, position sensing through built-in GPS system, requires no computer, has wireless communication via Bluetooth and WiFi, and easy to use! LAMP can be used to amplify a few copies of DNA to 109 copies in as little as 30 minutes.
The isothermal amplification Mastermixes allows fluorescence detection of the product on the Genie® platform but may also be used on generic qPCR instrumentation. An anneal curve can then be generated to confirm the product. This eliminates the need for gel electrophoresis or turbidity detection and allows for a closed-tube system.
Isothermal Mastermixes are fully licensed for LAMP use by Eiken Chemical Company. This Isothermal Mastermix can be used with RCA, SMAP and many other amplification technologies.
The ISO-001 master mix uses an engineered LF DNA polymerase isolated from Geobacillus sp. SSD , GspSSD. The enzyme shows highest speed in a fluorescent LAMP reaction and is the enzyme of choice for LAMP reactions.
Also available is a wide range of accessories which go with the Genie® instruments. These include reactions tubes, cooling blocks (also act as holders for the tubes), and robust carry case for your instrument.
